Webinar: High coverage single-cell metagenomics using PTA
A webinar, High coverage single-cell metagenomics using PTA, invited by Techtum and Bioskryb, is now uploaded!
The talk presents our approach to sequencing individual bacterial genomes using semi-permeable capsules and Primary Template-directed Amplification (PTA). These capsules retain genetic material while allowing reagents to be exchanged, enabling multi-step library preparation at scale.
Using data from more than 50,000 bacterial cells from a mock community and human saliva, we compare PTA with Multiple Displacement Amplification (MDA) and examine how cell lysis affects which bacteria are recovered. The talk also introduces our Bascet/Zorn analysis pipeline for single-cell whole-genome amplification data.
